Case study:
Context: A healthy gut is nowadays connected with a well-functioning body at systemic level. A classical approach to modulate the gut microbiota activity and composition is the use of prebiotics. A functional foods company wants to develop a prebiotic mix that can be consumed to exert a positive activity (i.e. sustained fermentation) along the entire colon. They have several lead compounds and they contacted you to understand which product is the most promising one. As a bioengineer you are involved in the development of this prebiotic product and you are asked to guide the different steps in the research process. Answer each question clearly using bullet points.
- Which type of fibers would you select to obtain a sustained fermentation along the entire colon? Motivate substantially.
- How can prebiotics exert a positive effect in the gut?
- Provide a short experimental plan to evaluate the effect of prebiotics making use of an in vitro preclinical platform.
- In the experimental design, would you consider it important to assess interindividual variability? Why is this parameter important?
- Which molecular quantification analyses would you use to assess the modulation of the gut microbiota composition in the presence of the prebiotic compounds?
- Which in vitro model (i.e. cell lines / organoids) would you use to assess the impact of the prebiotic at the level of the gut wall? What would you measure? Is the observed effect direct or indirect?
- Once you have selected the most interesting product, the client would like to assess host parameters making use of an animal model. Which options do you consider most promising for this study?

Studententip: Er werd niet echt zwaar in detail gevraagd en veel meer dan hier staat vroeg hij niet.
- Genetic susceptibility van IBD:
- Vertellen over leucine-rich repeats en de locus (vooral daar de mutaties) en dat dit een NOD is.
- De drie mechanismen benoemen (hoeft niet in detail; bijv. macrofagen kunnen het niet doden is al voldoende).
- MHC:
- Afkomstig uit het ER, presenteert antigenen, costimulerend signaal.
- Verschil exogeen vs. endogeen goed kennen (MHC I presenteert endogene antigenen).
- Bijvraag: Als een coronavirus in je cel komt, via welke MHC-klasse wordt dit gepresenteerd?

- Definitie geven en dysbiose uitleggen.
- Belang van keystone species i.p.v. per se hoge diversiteit.
